Phosphodiester bond at 5’ phosphate and 3’ hydroxyl group of adjacent nucleotides is joined by T4 DNA ligase in presence of ATP as a cofactor. It can be used for both blunt and sticky end in DNA cut. It bonds the nucleotides in double stranded DNA/RNA only.
